Abstract: | Cannabidiol (CBD) is a group of non-psychological components of Cannabis sativa that is widely used in medicine. CBD was demonstrated to exert its anti-inflammatory effect in distinct types of oral cells; however, its proliferative effect has not yet been extrapolated. Besides alleviation of periodontal disease progression, promotion of periodontal regeneration by enhanced proliferation of various cells within periodontium is another goal of periodontal therapy. This study aimed to determine the cytotoxicity of CBD and the proliferation of human gingival fibroblasts (HGFs) and periodontal ligament (PDL) cells upon treatment with CBD. Method: HGFs and PDL cells were treated with 0.01-30 µM of CBD or 1% methanol as a solvent control. An MTT assay was conducted for determination of cytotoxicity. The proliferative effect of CBD was investigated by a BrdU assay, and then confirmed by Western blot hybridization and immunofluorescence for expressions of cyclin D1 and Ki-67, two markers of cell proliferation. Result: The results showed no cytotoxicity was found in HGFs or PDL cells upon treatment with any doses of CBD up to 30 µM. The mean percentage of proliferation in HGFs was significantly enhanced by 3 and 10 µM of CBD (p<0.001), while that of PDL cells was found to be significantly increased by CBD as low as 0.01 µM up to 3 µM (p<0.01). Consistently, protein expressions of cyclin D1 and Ki-67 were up-regulated upon treatment with CBD from 1 to 10 µM. Conclusion: Treatment with non-toxic doses of CBD promotes the proliferation of HGFs and PDL cells in vitro. |
