Abstract: The pressure sensitive adhesive is a type of non-reactive adhesive generating a bonding as pressure is applied to bond the adhesive with a target surface. The present study prepared the clotrimazole-loaded in situ forming film (ISF) oral solutions comprising 40% or 50% of fatty acid mixture (1:1 lauric and palmitic acid) as matrix film forming agents and Duro-Tak® as pressure sensitive adhesive. Their physical properties including matrix formation, spray pattern, adhesiveness and antifungal properties were investigated. ISF oral solutions exhibited low density and viscosity and that comprising lesser amount of. Duro-tak® (2.5%) with 40% fatty acid mixture gave the wider spray angle. The ISF solutions containing 40 % and 50% fatty acid mixture could transform into matrix whereas the latter transformed into the strong matrix film at the interface between ISF system and agarose gel. The addition of 2.5% Duro-Tak® was suitable to enhance the adhesion of ISF on buccal and tongue tissues. Moreover, the prepared systems showed the inhibition against the Candida albicans. Thus, the developed clotrimazole-loaded ISF oral solution comprising 2.5% Duro-Tak® with blend of 40% fatty acid mixture exhibited the potential for treatment of oropharyngeal candidiasis.
