Abstract: Four species of Habenaria and 2 species of Pecteilis were employed in morphology study as well as genetic relationships. A total of 9 species and varieties was recorded each specific characteristics. Random Amplified Polymorphic DNA (RAPD) technique was used to analyze genetic relationships of those plant groups. DNAs were extracted using four different extraction buffers, CTAB (Cetyl trimethylammonium bromide), CTAB + 1% PVPP, SDS (Sodium dodecyl sulfate) and SDS + 1% PVPP buffers. It was shown that SDS buffer supplemented with 1% PVPP could yield better quality of DNAs. Dendrogram from 15 primer combinations for genetic relationship divided 9 plant samples into 2 groups. The first group was including a group of H. rhodocheila and H. xanthocheila. The second group was including H. lindleyana, H. myriotricha and 2 species of Pecteilis corresponded with common character as a colored flower and white flower, respectively. Specific markers could generate by OPA10680 in H. rhodocheila and H. xanthocheila and OPA10362 in H. lindleyana, H. myriotricha and 2 species of Pecteilis.
