Abstract: Endoplasmic reticulum (ER) stress, a commonly causes of neuronal damage involved with many diseases, including Alzheimer’s disease and Parkinson’s disease. Therefore, interventions that attenuate ER stress may contribute to reduction in apoptotic cell death. In this study, we examined the effect of O-demethyldemethoxycurcumin, curcumin analog in against thapsigargin (TG) - induced ER stress in SK-N-SH cells. In the results showed that O-demethyldemethoxycurcumin reduced SK-N-SH cells death in a concentration-dependent manner, decreased the number of cell apoptosis via attenuated intracellular Ca2+ and down regulated ER stress signaling proteins expression (p-PERK, p-IRE1, BiP, CPS12 and CHOP). Our findings suggest that O-demethyldemethoxycurcumin could protect against ER stress-induced SK-N-SH cells death induced by TG. Therefore, the potential of O-demethyldemethoxycurcumin may be one of alternative therapeutic agent for protective of neurodegenerative diseases
