Abstract: | Dental radiographs or X-rays have been numerously used in dentistry. It is known that the ionizing properties could damage human absorbing tissue though applying in the minimal amount. Salivary gland tissue cells are commonly exposed to radiation via the direction of dental radiography. This study was aimed to investigate the proliferative and apoptotic effects of dental irradiation on human salivary ductal cell lines in vitro. Two ductal cell lines, HSG and HSY, were characterized by carcinoembryonic antigen (CEA) and lactoferrin (LF) expressions, and examined for their doubling times. They were exposed to periapical radiography for 5, 10 or 20 times (1.1 mGy each) and further incubated for 1, 3 or 5 cycles of their cell division. Cell proliferation was examined by a BrdU assay and an immunoblot analysis for Ki- 67 and proliferating cell nuclear antigen (PCNA) expression. The apoptotic potential was determined by the ratio of B-cell lymphoma 2 (BCL-2)/BCL-2-associated X protein (BAX). |
