Abstract: | Objective: This study aimed to investigate the antifungal effect against Candida albicans (ATCC® 10231) and the durability of chitosan oligosaccharide (COS) coated on heat-cured polymethylmethacrylate (PMMA) surface. Material and method: Heat-cured PMMA specimens were manufactured. The various concentrations of COS solution; 0.5 MFC, 1 MFC and 2 MFC, were prepared and applied on specimens treated surface with three different methods: sandblasting (SB), application of 3-aminopropyltriethoxysilane (APS), and combined both methods (SB&APS). The toothbrush abrasion test was done to evaluate the percentage of remaining COS. The antifungal effect was determined through a microbiological assay using specimen preparations of the groups with favorable results. A number of fungal colonies were recorded at 16 and 48 hours. Data were analyzed using two-way and one-way ANOVA (p < 0.05) Results: COS concentration and surface treatment affected coating durability. The groups that applied APS revealed a high percentage of COS and significantly differed from those that used only sandblasting. Coatings with 1 MFC and 2 MFC demonstrate no difference, but coatings with 0.5 MFC differ significantly from others. At 16 and 48 hours, the specimens coated with 1 MFC and 2 MFC by APS and SB&APS methods inhibited the growth of C.albicans while the uncoated group (control) demonstrate no inhibition. Conclusion: By using APS, COS can be coated onto PMMA. Coated PMMA inhibited the growth of C.albicans. With further development, this method could be applied to treat denture stomatitis alternatively to conventional antifungal drugs, which lead to drug resistance and taste alterations. |
