Abstract: | This research exhibited that soybean (var. CM. 60) seed protein (3.00 µg/µL protein) was able to inhibit mushroom tyrosinase toward both monophenolase (32.86%) and diphenolase (43.30%). ABTS radical was decreased by 9.30-90.12%, when using 0.016-0.50 µg/µL protein. NO in RAW 264.7 cells was reduced by 10.89-47.77%, when using 0.19-1.50 µg/µL protein. Additionally, a protection toward H2O2-induced HaCaT cell oxidation was also found. ABTSradical scavenging activity and the protection toward H2O2 were contributed by small proteins (<10 kDa) more than large proteins (>10 kDa). The potent protection of <10 kDa (1.50 µg/µg protein) against H2O2 induced HaCaT cell oxidation was observed by increasing the cell viability to be 46.67%, resulting from the decrease of ROS and cell apoptosis. Moreover, CM. 60 protein showed low cytotoxicity toward HaCaT cells and Vero cells. These results suggest that soybean protein could be developed as a safe and potent skin whitening agent.
